L'edizione 2009-2010 della Premier League è stato il 111º massimo campionato inglese di calcio, nonché diciottesima edizione della Premier League. Ha aperto i battenti il 15 agosto 2009 e si è conclusa il 9 maggio 2010 con la vittoria del Chelsea.

Il campione in carica è il Manchester United. Le tre neopromosse sono Birmingham, Burnley e Wolverhampton, che rimpiazzano le retrocesse Newcastle, Middlesbrough e West Bromwich Albion.
